CyTOF & High-Dimensional Cytometry Core

The Georgia Cancer Center CyTOF & High-Dimensional Cytometry Core (RRID: SCR_025747) bridges the gap between the cutting-edge flow cytometry technologies and your research goals.

We provide end-to-end solutions from experimental and panel design to advanced high-dimensional data analysis.

Mass cytometry, or CyTOF (cytometry by time-of-flight), integrates the principles of flow cytometry with the quantitative capabilities of mass spectrometry, enabling multi-parametric single-cell analysis with exceptional throughput and resolution. By employing metal isotopes for antibody labeling, mass cytometry theoretically provides up to 140 detection channels with minimal signal spillover between channels.

CyTOF XTCyTOF-XT from Multiplex Bio
(formerly Standard Biotools)
has been in operation at the core since 2022.

Mass Cyto vs. Fluro image

FLUORESCENCE FLOW CYTOMETRY
VS.
MASS CYTOMETRY (CyTOF)
Broad, overlapping emission spectra  Signal Generation Discrete metal isotope masses
Compensation or spectral unmixing required Data Processing No compensation
Spillover spreading error Data Quality Minimal channel overlap
Affected by autofluorescence Background No autofluorescence
Tandem dye instability Reagent Stability Stable metal isotopes
Limited multiplexing (10-45 parameters typical) Multiplexing Capacity 50+ markers routinely
Complex panel design and optimization Panel Design More straightforward panel design
Live-cell sorting possible  Cell Recovery Cells are destroyed during analysis
Fast acquisition (high event rate) Acquisition Speed Slower acquisition (lower event rate)
